Working with DNA2.0We offer two mechanisms for accessing our ProteinGPS™ technology:1) Design-synthesis-analysis. In collaboration with a customer, we - Design and synthesize variant genes
- Transfer variants to the customer for protein expression and commercially relevant functional measurements
- Use customer-provided data to perform sequence-function analysis and design higher performing variants
2) Data-mining consultation. We also offer a sequence-function analysis for data that our customers already possess, for example from tiered library screening. Our customers perform all variant screeningOnce we have designed and synthesized a set of variants (generally between 20 and 100), the genes are transferred to our customers for measurement of activity. This approach serves several purposes. First, our customers are usually experts in measuring the properties of the target protein. We do not want to spend resources attempting to replicate existing capabilities. Second, we do not want to use surrogate or high-throughput screens. Such assays are frequently misleading and our aim is to optimize the target protein for a commercial purpose, not for a function in a high-throughput screen. Third, by focusing on what we do best and letting our customers do what they do best, we can work better and more cost effectively. The process is iterativeWhen we have defined a relationship between protein sequence and function, we use this information to design another small set of variants with improved performance under commmercial application conditions. This process may be repeated more than once, depending upon the degree of improvement required. ProteinGPS™ Workflow
